OXALIPLATIN: Oxaliptain exhibits a wide spectrum of both in vitro cytotoxicity and in vivo antitumour activity in a variety of tumour model systems including human colorectal concer models. Oxaliplatin also demonstrates in vitro and in vivo activity in varous cisplain resistant models. A synergistic cytotoxicaction has been obseved in cimbination with flurourocial both in vitor and in vivo. Studies on the mechnism of action of oxaliplatin although not completely elucidated, show that the aqua-derivatives resulting form the biotranstformation of oxalipation interact with DNA synthesis leading to cytotoxic and antitumour effects.

Indications: Adjuvant treatment of sate III (Duke’s C) colon cancer after complete resection of the primary tumour treatment of advanced colorectal cancer.

Dosage: In combination with fluorouracil and tolinic acid the recommened dose for the treatment of advance colorecal cancer is 85mg/m2 intravenously repeated every tow weeks or 130mg/m2 repeated every three weeks in combinatin with fluorouracil and foinic acid the recommended dose for adjuvatn treatment is 85mg/m2 intravenously repeated every two weeks for 12 cycles 96 months)

Contra Indication: Hypersensitivity to Oxaliplatin pregnatn breat feeding myelosuppression pior to starting first coures, as evidenced by beseline neutrophils, <1.5x109/l and/ neuropathy with functional ipairment pirio to first course, severly impaired renal function (Creatinine clearance less than 30mL/min).

Special Precautions: Oxaliplatin should be administered only by or under the supervision of an experienced cilincal oncologist not recommended in pregnancy, lactation & in children because of insufficiency data available.
