Fluconazole

Fluconazole is effective fungal infections. It is also used to treat yeast infections of the, mouth, throat, vagina, esophagus, lungs, abdomen, blood, and other parts of a human body. Fluconazole belongs to a class of antifungals known as triazoles. It slows the growth of fungi that are behind the infection. Fluconazole is available as a tablet and in liquid.

Fluconazole also prevents:

• - infection of the membranes covering the brain and spine)
• - to prevent yeast infections in people who may be infected because of chemotherapy or radiation therapy

The duration of the use of fluconazole depends on the condition of the patient and on how well the body responds to fluconazole.

Technical detail:

FLUCONAZOLE: It is a bis-triazole antifungal drug & is structurally related to imidazole-derivatives. It is fungistatic ain action & exerts its antifungal activity by altering cellular membranes resultin in increased membrane pemeability,leak-age of seential elements (e.g amino acids, potassuim) and impaired uptake of preursormolecules (e.g. purine & precusor to DNA).

Indications: Systemic candidiasis, mucosal candidiasis & cyptococcosis, vaginal candidiasis, fungal infections in patients with malignancy.

Dosage: Orpharyngeal or Oesophagel candidiasis, 200mg on the first day follwed by 100mg once daily. Max 400mg/day. Vanginal condidiasis: 150mg once oral dose. Deep setaed candidiasis : 400mg on first day then 200mg once daily for 4wks & for atleast 2 wks after resolution of symptoms. Cyptoccoccal menigitis: 400mg on first day followed by 200mg once daily Max. 400mg/day for 10-12wks. Theretafter 200mg once daily. Neutorpenica cancer : 50-100mg daily

Contraindications: Hypersensitivity

Special Precaution: Liver dysfunction, immunocompromised patients.

Pediatrics: Reduced dose necessary. Contraindicated in age below 4 weeks.
Pregnancy: May affect eht foetus.
Lactation: Drug passes int breast milk.

Side-effects: Nausea,headache, skin rash abdomina pain, vomiting & diarrhoea.

Drug interactions: Increases the serum level of phenytoin, warfarin and sulfonylureas.
