Leflunomide
Lefiunomide is an isoxazole immunomodulatory agent inhibits dihydrorotate dehydrogenase activity of the enzyme. It has immunosupressive and anti proliferative properties.
INDICATIONS : Rheumatoid arthritis
CONTRAINDICATIONS : Hypersensit vity, severe immunodeficiency, expecting woman.
SAFETY PROFILE : Monitor liver function regularly, renal insufficiency, bone marrow dysplasia, recent treatment with other hepatoxic or myelotoxic disease modifying anti rheumatic drugs.
ADVERSE EFFECTS : Abnormal liver enzymes, anorexia, paraesthesia, gastroenteritis, anxiety, hypokalemia, leukopenia, eosinophilia tendon rupture, dizziness, abdominal pain, diarrhoea, nausea, vomiting.
DRUG INTERACTIONS: Cholestyramine and charcoal decrease active metabolite. Rifampicin increase leftunmide plasma levels.
DOSAGE : Loading dose : 10 mg daily for 3 days Maintenance dose 20 mg/day.
