It has got central anti dopaminergic effect and thereby raises threshold for vomiting at chemoreceptor trigger zone. Its effects on the G.I. tract are thought to result from blocking of dopaminergic receptors; potentiation of cholingergic effects and/or a direct action on smooth muscle. Metoclopramide stimulates the release of prolactin.
INDICATIONS: Nausea and vomiting, gastritis, gastrooesophageal reflux, heart burn. Hiccups. Pre/post operative nausea, vomiting. Radiological examinations. Diagnostic procedures in gastroenterology. Lactation failure.
DOSAGE: 10 mg thrice daily children 0.5 mg/kg body wt per day in 3-4 divided doses.
CONTRA-INDICATIONS: Hypersensitivity, gastrointestinal haemorrhage, obstruction or perforation. Epilepsy or extra-pyramidal syndrome. Phaeochromocytoma, Breast carcinoma.
SIDE-EFFECTS: Restlessness, drowsiness, fatigue, lassitude, extra-pyramidal effects, tardive dyskinesia, motor restlessness, galactorrhoea & gynae comastia.
DRUG INTERACTIONS: Reduces absorption of oral digoxin. Increases absorption of aspirin paracetamol diazepam. Antagonsed by anti cholinergics antimuscarinic agents opioid analgesics.
