Powerful dopamine antagonist having anti emetic activity. It increases gastric emptying and shortens the stay of liquids, semisolids and solid in the stomach.
INDICATIONS: Acute nausea and vomiting, reflux oesophagitis. Dyspepsia, Peptic ulcer, uraemia.
CONTRAINDICATIONS: GI obstructions or perforation epilepsy, phaeochromocytoma, extra pyramidal symptoms.
SAFETY PROFILE: Beneficial effects of domperidone may be antagonized by anti cholinergic drugs. Renal impairment.
ADVERSE EFFECTS:Galactorrhoea in females and gynaecomastia in males due to increased production of prolactin.
DRUG INTERACTIONS: Digoxin: Decreased absorption of digoxin, aspirin, paracetamol and oral diazeapam: increased absorption of these drugs. Anti cholinergic drugs : Effect of domperidone antagonized. Phenothiazines : increased CNS depression.
DOSAGE: 10 -20 mg every 4-8 hours. Children 0.3 mg/kg body weight 3-4 times daily. Before meals and bed time.
