It is a proton pump inhibitor that suppresses gastric acid secretion by specific inhabitation of the H+ / K+ ATPase in the gastric parietal cell.
INDICATIONS: Gastroesophageal reflux disease. Eradication of Haelicobacter pylori in combine with appropriate antibacterial regimens. Healing and maintance of erosive oesophagitis.
CONTRAINDICATIONS: Hypersensitivity to any component of the formulation.
SAFETY PROFILE: Reduced dose in hepatic insufficiency. Should be taken one hour before meals.
ADVERSE EFFECTS: Headache, flatulence, dry mouth, alkaline phosphatase.
DRUG INTERACTIONS: Esomeprazole may interfare with the absorption of drugs where gastric PH is an important determinant of bioavailability e.g. ketokonazole, iron salts and digoxin.
DOSAGE: Gastroesophageal reflux disease, healing of erosive esophagitis, 20 mg or 40 mg once daily for 4 to 8 weaks. Maintenance of healing of erosive oesophagitis. 20 mg once daily. Symptomatic gastroesophageal reflux : 20 mg once daily for 4 weeks. H-pylori eradication to reduce the risk of duodenal with amoxicillin, 1 g twice daily and clarithromycin 500 mg twice daily for 10 days.
