Omeprazle is a gastric proton pump inhibitiors. This prodrug is a substituted benzimidazole derivative which acts on the final step of gastric acid production and therefore controls acid secretion in response to all varieties of stimuli, omeprazole has prolonged action in a dose of 20-40 mg once a day and reduces the 24 hours gastric acid output by almost 90%. It is highly effective in Zollinger-Ellison syndrome, recurrent duodenal ulcers & reflux oesophagitis. It also suppresses H-pylori. It should be taken before meals.
INDICATIONS: Duodenal & gastric ulcers, NSAIDs induced ulcers, Zollinger-Ellison syndrome,b reflux oesophagitis.
CONTRAINDICATIONS: Hypersensitivity, pregnancy, lactation, children.
SAFETY PROFILE: Exclude the gastric malignancy, renal & hepatic dysfunction, epilepsy, avoid its use with alcohol.
ADVERS EFFECTS: Skin rashes, abdominal colic, diarrhea, nausea, dry mouth, dizziness, headache, constipation & flatulence, increased risk of enteric infections due to reduced acid secretion.
DRUG INTERACTIONS: Diazepam, phenytoin, warfarin, cyclosporine & digoxin.
DOSAGE: Peptic ulcers-20 mg once daily for 4 weeks max up to 8 weeks.
Reflux oesophagitis-20 mg once daily for 4-8 weeks.
Z.E.S- 60 mg once daily to start than 20-120 mg daily in 2 divided dose.
