Ezetimibe is a class of lipid lowering compounds that selectively inhibits the intestinal absorption of cholesterol and related phytosterol. 3( R)[3-(4-fluorophenyl) -3(s)-hydroxypropyl]-4(S)-(4-hydroxyphenyl)-2azetidinone. The empirical formula is c24 H21 F2 NO3.
Ezetimibe is a white crystalline powder that is freely to very soluble in ethanol, methanol and acetone and practically insoluble in water. Ezetimibe has a melting point of about 163 C and is stable at ambient temperature. Ezetimibe is available as a tablet for oral administration containing 10 mg of ezitimibe.
INDICATIONS: Primary hyperchloesterolaemia Homozygous amilial hypercholesterolaemia, homozygous sitosterolaemia.
CONTRAINDICATIONS: The combination of ezetimibe with an HMG CoA reductase inhibitor is contraindicated in patients with active liver disease or unexplained persistent elevation in serum transaminases. Prior hypersensitivity of Ezetimibe.
SAFETY PROFILE: Liver function test should be done if co-administrated with statin. Should not prescribe to the patient with modereate or severe hepatie insufficiency.
ADVERSE EFFECTS: Diarrhoea, back pain, respiratory infection, fatique.
DRUG INTERACTIONS: Cholestyaramine, cydosporine.
DOSAGE : 10 mg once daily orally.
