Nimesulide
Non steroidal anti-inflammatory drug with minimal gastric side effects has some selectivity for Cox-2 than Cox-1. In addition to inhibition of prostaglandin synthesis. It also reduces generation of superoxide by neutrophils, inhibits PAF synthesis, Inhibits TNE and inhibits proteinase activity in cartilage, thus preventing breakdown of osteo-arthritic human cartilage.
Indications : Osteoarthritis, dysmenorrhoea, rheumatoid, arthritis, low back pain, dysmenorrhoea, dentalpain, post operative pain, trauma.
Contradictions : GI bleed, peptic ulcer, hepatic dysfunction, pregnancy, lactation.
Safety Profile : Reduce dose in elderly, renal dysfunction, heart failure.
Adverse Effects : Epigastralgia, heart burn, nausea, loose motions, rash, pruritus, somnolence, dizziness.
Dosage : 100 mg twice a day
Children : 5 mg/kg/day in two or three divided dose.
