Amoxapine
A tetracyclic antidepressant having less cardiovascular and anti cholinergic side effects.
Indications: Obsessive compulsive neurosis, phobic states, endogenous depression, for restoring normal social behaviour.
Contradictions: Renal, hepatic and cardiac insufficiency.
Safety Profile: Patients with diabetes, liver diseases, myocardial infarction Epilepsy and mania Elderly patients.
Adverse Effects: Drowsiness, jaundice, Blood dyscrasias, postural hypotension, Cardiac arrhythmias.
Drug Interactions: Alcohol: sedative effect increased. Anti Parkinson’s and anti psychotic drugs: Increase anti cholinergic activity.
Anti hypertensive drugs: Anti hypertensive activity reduced.
Barbiturates: Serum level of amoxapine reduced. Neuroleptics: Inhibit metabolism.
Dosage: Adults: start with 100-150 mg daily. Increase to 300mg daily in divided doses.
Elderly start with 50mg daily in divided doses. Increasing after 5-7 days to 50mg t.i.d.
