AMITRIPTYLINE
It blocks the neuronal up take of noradrenaline. It has marked effect in comparison to imipramine.
INDICATIONS : Depression associated with anxiety, agitation, restlessness and disturbances in sleep, bed wetting by children.
CONTRAINDICATIONS : PREGNANCY, liver dysfunctions, ischemic heart disease, epilepsy, glaucoma, lactation.
SAFETY PROFILE : Patients with history of hepatic, cardiac and renal diseases. Ability to drive or operate heavy machinery may be impaired. May induce suicidal tendency indepressed patients.
ADVERSE EFFECTS : Postural hypotension, palpitation, blurring of vision. Dryness of mouth, epigastric distress, constipation, sedation, confusion, tremors, lowered seizure threshold. Urinary retention.
DRUG INTERACTIONS : Like amoxapine.
DOSAGE : Adults : start with 75mg daily in divided doses. Increasing upto 150mg daily in divided dose, Elderly 10mg thrice daily.
Children : below 12 yrs. Not recommended.
