Trimipramine
More sedative in comparison to other tricyclic antidepressants. More useful in patients of depression with anxiety and agitation. Effect is seen after a latent period of 3 weeks.
Indications: Depression associated with sleep disturbances, restlessness, and depression in alcoholics.
Contradictions: Epilepsy, Ischemic heart disease, narrow angle glaucoma.
Safety Profile:
Adverse Effects: Dryness of mouth, constipation, epigastric distress, urinary retention, blurring of vision, tremors, convulsions, confusion.
Drug Interactions: Guanethidine, methyl dopa: anti hypertensive action impaired. MAOI: Hyperpyrexia, convulsions, death. Alcohol : potentiates CNS effects.
Dosage: 50-100 mg. given 2 hours before bed time. Moderate/severe depression (Hospitalised patients) 75mg as a single dose or in divided doses, gradually increasing to 150-300 mg daily. Continue for 4-6 weeks then gradually reduce to maintenance dose.
